Endosperm culture

For culture of mature endosperm, fruits are
surface sterilized and endosperm plus embryo
are dissected out under aseptic conditions.

For culture of immature endosperm, entire
seeds are sterilized and the endosperm tissue
is carefully excised under aseptic conditions
using a stereoscopic microscope. It is desirable
to dissecting out endosperm explants free
of embryo tissue. Failure in this regard could
lead to contamination of the endosperm callus
with the tissue of embryo origin (diploid tissue).
